PROBLEM TO BE SOLVED: To provide a method for the separation and purification of a nucleic acid containing a step to adsorb the nucleic acid to a solid phase by pressurizing a nucleic acid containing specimen solution in a container and passing the specimen solution through the solid phase placed in the container while preventing the formation of droplets of the liquid in the container from leaving the droplet liquid behind in the container. SOLUTION: The method for the separation and purification of a nucleic acid contains (1) a step to adsorb the nucleic acid to the solid phase by pressurizing a nucleic acid containing specimen solution in the container and passing the solution through the solid phase placed in the container, (2) a step to add a cleaning liquid to the container and pressurize the cleaning liquid to pass the liquid through the solid phase, and (3) a step to add a liquid for desorbing the nucleic acid from the solid phase to the container, pressurize the liquid and pass through the solid phase to recover the nucleic acid in the liquid.
