<P>PROBLEM TO BE SOLVED: To carry out measuring concentration of a material to be inspected with high sensitivity, without eluting electron acceptors into an aqueous sample, by using an enzyme electrode of an electron mediator-type biosensor. <P>SOLUTION: In the biosensor, an enzyme immobilizing membrane of a working electrode is made up of two components which are an enzyme reaction layer and a charge transfer medium layer. The enzyme reaction layer is made up so that oxidoreductase is fixed and held in a hydrophilic macromolecule medium layer, and the charge transfer medium layer is made up so that the electron acceptor is held in a hydrophobic macromolecule layer. The electron acceptor under a hydrophobic environment is not eluted easily into the aqueous sample. Therefore, the concentration measurement of the material to be inspected can be carried out with high sensitivity and high precision, and the measurement sensitivity is not decreased even in the case the electrode is used repeatedly, and an accurate calibration can be carried out when using calibration liquid for the calibration because no electron acceptor is eluted.
