The present invention is directed to a novel targeted vaccine delivery system, comprising one or more isolated MHC class I alpha3 domains linked to an antibody which is specific for a cell surface marker and associated with beta2-microglobulin and antigenic peptides, or costimulatory molecules, or cytokines. The complexes of the invention are useful for treating and/or preventing cancer, infectious diseases, autoimmune diseases, and/or allergies.
