PROBLEM TO BE SOLVED: To provide a method for detecting a specific base sequence of one DNA molecule. SOLUTION: The method for detecting the specific base sequence of the one DNA molecule comprises forming a composite monomolecular membrane comprising a cationic amphipathic material and the DNA sequence-specifically bonded to a protein on a gas-liquid interface, and transferring the composite monomolecular membrane onto a solid substrate to elongate and immobilize the DNA on the solid substrate. The method enables the specific base sequence of the one DNA molecule to be easily detected by elongating and immobilizing the DNA bonded to the protein in good repeatability and good efficiency. Both of a fluorescent image and an image of appearance can be observed and measured by using a nearfield optical microscope to enable the accuracy to be improved. COPYRIGHT: (C)2005,JPO&NCIPI
