METHOD OF SCREENING SUBSTANCE INTERACTING WITH ABC PROTEIN

It is intended to provide a method of assaying the transporter activity of an ABC protein whereby a large number of samples can be simultaneously and conveniently assayed with little data variation at a high reproducibility even in the case of using the samples in a small amount; a method of screening a substance interacting with an ABC protein with the use of the above assay method; and a kit for assaying the transporter activity of an ABC protein which is advantageously usable in these methods. Preincubation is carried out by adding insect cell membrane expressing human P glycoprotein to a reaction buffer solution containing vanadic acid and [<3>H] ATP. Next, a test compound is added thereto followed by reaction. After the completion of the reaction, the liquid reaction mixture is added to a 96-well glass filter and the [<3>H] ATP adsorbed to the P glycoprotein-expressing membrane is quantified by the suction washing method in a single step.
