摘要 PURPOSE: A microsphere composition of suspension array for analyzing Hepatitis C virus (HCV) genotype and a detecting method thereof are provided, thereby rapidly and accurately analyzing Hepatitis C virus (HCV) genotype, and analyzing HCV la to 4a genotypes. CONSTITUTION: The oligonucleotide probe composition of suspension array for analyzing Hepatitis C virus (HCV) genotype comprises the nucleotide sequences set forth in SEQ ID NO:1 to SEQ ID NO:15, wherein the oligonucleotide probe is immobilized onto the microsphere; a first or second primer sequence for detecting the binding of the probe with the target gene has the nucleotide sequence set forth in SEQ ID NO:16 to SEQ ID NO:19; and the anti-sense primer of SEQ ID NO:19 in the second primer contains biotin. The method for detecting the Hepatitis C virus(HCV) genotype comprises the steps of: (a) extracting HCV RNA from blood plasma and serum; (b) RT-PCR amplifying the extracted RNA; (c) asymmetric PCR amplifying the RT-PCR amplified product; (d) binding the asymmetric PCR amplified product with the probe; and (e) verifying the binding of the asymmetric PCR amplified product with the probe by simultaneously reacting Streptavidin-Cy3 or Streptavidin-phycoerythrin which is capable of binding with biotin and verifying the reaction by using Luminex 100 LabMAP.
