<P>PROBLEM TO BE SOLVED: To provide a method for amplifying DNA high in amplification efficiency and easy in primer design. <P>SOLUTION: The method of amplifying DNA by PCR using sense primer and anti-sense primer comprises carrying out the PCR in the presence of an additional sense primer where an oligodeoxyribonucleotide having a 1st addition sequence is joined to the 5'-terminal of a sense primer and an additional anti-sense primer where an oligodeoxyribonucleotide having a 2nd addition sequence complementary to the 1st addition sequence is joined to the 5'-terminal of an anti-sense primer, wherein the Tm of the 1st or 2nd addition sequence is lower than the Tm of each of the sense primer and anti-sense primer, and the annealing temperature in the PCR is set at such a level as not to anneal the above addition sequence at the initial stage and changed to such a level as to anneal the sequence at a midway stage. <P>COPYRIGHT: (C)2004,JPO&NCIPI |
