摘要 <P>PROBLEM TO BE SOLVED: To provide a peptide C terminus amino acid sequence analysis method utilizing a decomposition reaction means for successive C terminus amino acid, restraining unfavorable secondary reaction such as cutting of peptide bond in the middle of peptide to perform the processing under very versatile conditions in sequentially decomposing the peptide C terminus amino acid with a long amio acid length. <P>SOLUTION: Dried sample of long peptide with a long amino acid is previously subjected to N-acylation processing. C terminus amino acid is decomposed under mild conditions by use of integrated reaction reagent obtained by combination of alkanoic acid anhydride and a small quantity of perfluoro alkanoic acid, subjected to hydrolysis, and selectively fragmentated at arginine residue part by trypsin digestion. After that, a decrease in molecular weight at the C terminus fragment derived from a series of reaction product is measured by a MALDI-TOF-MS device to specify the C terminus amino acid sequence. <P>COPYRIGHT: (C)2004,JPO&NCIPI
