摘要 PURPOSE: An hTERT(human telomere reverse transcriptase) promoter with enhanced tumor-specificity and strength and a recombinant vector comprising the same are provided, thereby specifically and highly expressing the hTERT gene in only cancer cells. Therefore, cancer cells can be selectively killed. CONSTITUTION: A transcription regulating sequence comprises the hTERT promoter and one or more of c-Myc binding sites and/or one or more of Sp1 binding sites which are linked to the hTERT promoter, wherein the c-Myc binding site has the nucleotide sequence selected from SEQ ID NO: 2, SEQ ID NO:3 and SEQ ID NO:4; the Sp1 binding site has the nucleotide sequence selected from SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:7, SEQ ID NO:8, SEQ ID NO:9, SEQ ID NO:10 and SEQ ID NO:11; the regulating sequence has the nucleotide sequence set forth in SEQ ID NO:14. The recombinant vector comprises the transcription regulating sequence which is operably linked to the gene required for virus replication, wherein the virus vector is derived from Adenoviridiae; the gene required for virus replication is an initial gene E1A.
