FIELD: biotechnology, pharmacy, preparative biochemistry. ^ SUBSTANCE: invention proposes a method for preparing the preparation interleukin-1-beta from biomass of recombinant strains of Escherichia coli. Bacterial cells are disrupted by treatment with alkali at pH 12-13, cellular walls are separated by centrifugation, proteins are precipitated by treatment of supernatant with ammonium sulfate and prepared precipitate is dissolved in tris-buffer. Prepared solution is subjected for three-stage chromatography purification. The first stage involves chromatography on cation-exchange resin Soloza KG and Q-cellulose. Then purification is carried out on hydrophobic sorbent Spherocell-Octyl at pH 3.9 using the solution containing 0.05 M tris-(hydroxymethylamino)methane, 0.05 M of sodium chloride and 0.0125 M of acetic acid as an eluent. Obtained mixture is concentrated on cationic-exchange resin Soloza KG. Invention provides preparing apyrogenic interleukin-1-beta with the enhanced yield. ^ EFFECT: improved preparing method. ^ 3 cl |
