VECTORS FOR EXPRESSION OF TOXINS

The invention relates to a method of manufacturing vectors encoding recombinant proteins or glycoproteins that are toxic to cells in which they are expressed. As a result, expression of these proteins must be tightly controlled to allow, for example, production of viral vectors for gene therapy in cells that would otherwise be killed by the toxin encoded by the vectors. Vectors comprising highly tissue-specific synthetic promoter constructs are described, which allow expression of the toxin only in cells having a nuclear accumulation of beta-catenin.
