BETA-AMYLOID BINDING FACTORS AND INHIBITORS THEREOF

PURPOSE: Beta-amyloid binding factors and inhibitors thereof are provided, which beta-amyloid binding factors is VEGF(vascular endothelial growth factor) polypeptide and can be useful for diagnosis and treatment of Alzheimer's disease. CONSTITUTION: The beta-amyloid binding polypeptide(β-ABP) specifically binds heparin and β-amyloid, wherein the polypeptide is not SEQ ID NO:5, the beta-amyloid binding polypeptide(β-ABP) has at least 85%, 90%, 95%, and 97% sequence identity to SEQ ID NO:3; the beta-amyloid binding polypeptide(β-ABP) is represented by SEQ ID NO:3, and may have added or subtracted 5 amino acids from either the N-terminal or C-terminal end; the beta-amyloid binding polypeptide(β-ABP) specifically binds to a region on β-amyloid that is from Gly residue at position 25 to Met residue at position 35 of SEQ ID NO:4; and the beta-amyloid binding polypeptide(β-ABP) has a binding affinity for β-amyloid represented by a dissociation constant of 10 nM, 1 nM or 100 pM. A method of preventing binding between β-amyloid and endogenous VEGF comprises the steps of: (a) generating a recombinant viral or plasmid vector comprising a DNA sequence encoding VEGF polypeptide operatively linked to a promoter; and (b) administering the viral or plasmid vector to a patient in need thereof, such that expression of the DNA sequence within a brain results in binding between β-amyloid and VEGF polypeptide.
