摘要 <P>PROBLEM TO BE SOLVED: To provide a method for increasing a number of copies of a specific nucleic acid sequence, i.e., "target sequence" which may exist singly, exist as a homogeneous component of a nucleic acid, or exist as components (in a large amount or a small amount) of a heterogeneous mixture thereof. <P>SOLUTION: In this method, two or more RNA copies of the target sequence further form copies in an autolytic manner for the purpose of initiating DNA and RNA syntheses by using a mixture of a blocked primer, a non-blocked primer, and/or a promoter-primer, so that two or more copies of the target nucleic acid sequence are synthesized in the autolytic manner under conditions in which formation of nonspecific products is preferably made to be reduced and of which the temperature, the ionic strength, and the pH are kept at substantially constant. Further, a composition for synthesizing the copies and a kit for the same are provided, respectively. An alkane-diol expressed by the figure in the specification is used as a blocking agent or a modifying agent. The method is useful for forming the copies of the nucleic acid target sequence, for the purposes of conducting an assay in which the specific nucleic acid sequence in a clinical sample, an environmental sample, a legal sample or the like is quantitatively analyzed, of cloning the same, and of forming a probe. <P>COPYRIGHT: (C)2004,JPO&NCIPI
