摘要 PROBLEM TO BE SOLVED: To provide a method for decreasing a non-specific reaction (non-specific hybrid forming, non-specific extension reaction) and improving the specificity of an enzymatic reaction. SOLUTION: The subject enzymatic reaction method uses a nucleic acid hybrid as a substrate and is provided with a step to react a target nucleic acid with a hybrid-forming nucleic acid having a sequence complementary to the target sequence of the target nucleic acid and forming a hybrid between the nucleic acids. In the hybrid-forming step, a competitive nucleic acid competing with the hybrid-forming nucleic acid is added in addition to the target nucleic acid and the hybrid-forming nucleic acid; the competitive nucleic acid has a sequence completely complementary to a mismatch sequence of the target nucleic acid, possible to form a mismatch hybrid by the hybrid-forming nucleic acid; and the 5'-terminal and/or the 3'-terminal of the competitive nucleic acid is chemically modified to inhibit the enzymatic reaction.
