发明名称 Beta-catenin oligonucleotide michrochip and method for detecting beta-catenin mutations employing same
摘要 <p>A beta -catenin oligonucleotide microchip for detecting beta -catenin mutations comprises oligonucleotides fixed on the surface of a solid matrix. The oligonucleotides are designed to detect a variety of mutations at mutational hot spots of the beta -catenin gene. Independent claims are also included for: (1) manufacture of the beta -catenin oligonucleotide microchip comprising: (a) mixing each oligonucleotide in a micro spotting solution and distributing to a well plate; (b) spotting the oligonucleotides on the surface of a solid matrix using a microarrayer; (c) fixing the oligonucleotides on the solid matrix surface and washing; (d) denaturing the fixed oligonucleotides by soaking the solid matrix in 95 [deg]C water and then treating the solid matrix with a sodium borohydride solution; and (e) washing and drying the solid matrix; and (2) detecting a beta -catenin mutation using the microchip, comprising: (a) preparing a fluorescent dye-labeled DNA sample from the blood of a subject; (b) reacting the labeled DNA sample with oligonucleotide spots on the beta-catenin oligo chip; (c) washing the reacted oligo chip to remove unbound sample DNA; (d) detecting the mode of hybridization of specific oligonucleotide spots using a fluorescence reader; and (e) examining the presence of gene mutation.</p>
