摘要 PROBLEM TO BE SOLVED: To provide a method for quickly and easily determining a base sequence at a polymorphic site in a target polynucleotide sequence. SOLUTION: This method for detecting the polymorphism of a target sequence in a nucleic acid sample comprises the step of hybridizing the nucleic acid sample with a plurality of probes hybridizable with the target sequence in the nucleic acid sample and comprising a 1st probe having on its end a sequence complementary to the polymorphic site of the target sequence and a 2nd probe adjoining the 1st probe at their ends and hybridizable with a sequence adjoining the polymorphic site, wherein the 1st and/or the 2nd probe(s) is(are) fluorescent-labeled, the step of carrying out a ligation of the 1st and 2nd probes hybridized with the target sequence, and the step of dissociating the ligation reaction product from the target sequence. The method is performed by determining the target sequence by detecting the fluctuation of the fluorescence intensity of the fluorescent-labeled 1st and 2nd probes and discriminating presence/absence of ligation by fluorescence spectroscopy.
