METHOD OF ANALYZING C-TERMINAL AMINO ACID SEQUENCE OF PEPTIDE

It is intended to provide a method of analyzing the C-terminal amino acid sequence of a peptide with the use of a means of successively degrading C-terminal amino acids whereby the C-terminal amino acids of a peptide having a long amino acid chain can be successively degraded while preventing undesirable side-reactions such as cleavage of a peptide chain in the middle of the peptide and the treatment can be carried out under widely applicable conditions. Namely, a method of specifying the C-terminal amino acid sequence by preliminarily N-acylating a dry sample of a peptide having a long amino acid chain, successively degrading the C-terminal amino acids under mild conditions by using a reaction reagent comprising an alkanoic acid anhydride combined with a small amount of a perfluoroalkanoic acid, hydrolyzing, then digesting the obtained product with trypsin to thereby selectively fractionate it at an arginine site, and then measuring a molecular weight loss of the C-terminal fragment caused by a series of the reaction products by the negative mode analysis with the use of an MALDI-TOF-MS apparatus.
