METHOD OF CONSTRUCTING ASSIGNED MOLECULE AND LIBRARY COMPRISING THE SAME AND METHOD OF USING THE SAME

An assigned molecule cDNA library is constructed by forming a single-stranded DNA by reverse transcription with the use of a random primer from an RNA or mRNA library; using the single-stranded DNA as a template, constructing a double-stranded DNA library phosphorylated exclusively at the 5'-end of the complementary strand DNA of the single-stranded DNA; ligating the double-stranded DNA library with an adaptor made of a nucleic acid not phosphorylated at the end to give a ligated double-stranded DNA library; using the ligated double-stranded DNA library as a template, constructing an assigned molecule cDNA library by PCR with the use of a primer having a sequence required in forming an assigned molecule.
