FIELD: medicinal microbiology. ^ SUBSTANCE: invention can be used for investigation of antigenic structure of anthrax microbe and identification of different genotypes in Bacillus anthracis. High-molecular somatic antigen is isolated from cultural filtrate of the plasmid-free strain B. anthracis 81/1 (tox-, cap-) that is cultured in R-medium with shuttling at 37 C for 22 h. Additional purification of somatic antigen is carried out by separation of cultural filtrate by gel-chromatography method on superfine Sephacryl S-300. Somatic antigen (molecular mass is above 150 kDa) comprises in the fraction I that is eluted as free volume (i. e. in the volume of eluting blue dextran) in recording the eluting fraction at lambda = 280 nm. Antigen doesn't induce skin edema in intracutaneous administration in guinea pigs in the dose 0.1 ml with the protein concentration 75 mcg/ml and it doesn't provide protection of animals in intramuscular administration of 100 and 150 mcg of adjuvant with incomplete Freund's adjuvant in 2 weeks and infection of animals with the highly virulent strain of B. anthrax 81/1 in 21 day. The prepared somatic antigen can be used for the more full study of antigenic properties of different genotypes in B. anthracis. ^ EFFECT: improved isolating method, valuable biological and medicinal properties of antigen. ^ 2 dwg, 1 tbl, 6 ex |
