METHOD FOR PREPARING IMMUNE MEMBRANE STRIP USING AVIDIN- OR STREPTAVIDIN-BIOTIN REACTION

PURPOSE: A method for preparing an immune membrane strip using avidin- or streptavidin-biotin reaction is provided, thereby improving sensitivity of the membrane strip, and effectively immobilizing antibody on the surface of the membrane strip. CONSTITUTION: The method for preparing an immune membrane strip using avidin- or streptavidin-biotin reaction comprises the steps of: immobilizing a biotin-unreactive protein polymer on a portion of the surface of the membrane; treating the remained portion of the surface of the membrane with nonreactive protein; transferring the streptavidin or avidin solution to the biotin protein immobilized area to induce a first (strept)avidin-biotin reaction; sequentially transferring biotin-antibody polymer solution into the same area to induce a second (strept)avidin-biotin reaction in order to immobilize the antibody; and washing the membrane strip, wherein the biotin-unreactive protein polymer is synthesized from protein which does not participate in a certain antigen-antibody reaction, such as biotin and bovine serum albumin having functional group, casein or gelatin; the number of biotin reacted with one protein is 2 or more; the membrane is synthetic resin having the pore size of 0.1 to 20 micrometer such as nitrocellulose, glass fiber or nylon and polyvinylidene fluoride(PVDF).
