The invention concerns a physico-chemical method for analyzing and sequencing molecules in a sample, said method using a spectrometer and a light beam of molecular thickness, called molecular spotlight for measuring transmission, diffusion and emission. The emission is stimulated so as to reduce the measurement time and to locate the emitting sequence position. This requires an intensive cooling device provided with several laser beams, fixed contacts for evacuating heat and fast flowing fluids, the quantum yield of the detection being enhanced by a special sensing and deflecting optical system. Maximum cooling of the amplifying optical system enhances the signal-to-noise ratio. The main fields of application of the invention are the key technology of molecular diagnosis, reading highly-compact stored data and polymeric analysis for exact determination tacticity and length of block copolymers and for measuring atomic-molecular progression lengths, even for macroscopic distances. |
