It is intended to provide a method of detecting hybridization which comprises immobilizing a nucleic acid probe (for example, a single-stranded oligo DNA) on an electrode, adding a nucleic acid sample (for example, a specimen DNA) and then electrochemically measuring whether or not a double-strand such as a double-stranded DNA is formed, and by which the double-strand formation of a double-stranded DNA can be detected at a high accuracy without resort to any dye and the measurement can be carried out repeatedly. Namely, a method of detecting hybridization between a target nucleic acid and a nucleic acid probe which comprises allowing a single-stranded nucleic acid probe immobilized on an electrode surface to coexist with a nucleic acid sample having been denatured into a single-stranded one, and then detecting the hybridization of the nucleic acid probe with the target nucleic acid contained in the above-described nucleic acid sample by measuring the alternate current impedance of the electrode carrying the nucleic acid probe immobilized thereon. |
