Isolating biological modulators from biodiverse gene fragment libraries

A method of identifying nucleic acid that encodes a peptide having a conformation sufficient for binding to a target protein or nucleic acid. The peptide does not bind to the protein or nucleic acid in its native environment. The method comprises: a) constructing an expression library by a disclosed process; and b) screening expression library to identify a nucleic acid fragment that encodes a peptide that binds to the target protein or nucleic acid; and c) selecting a nucleic acid fragment at (b) that encodes a peptide that does not bind to target protein or nucleic acid in its native environment.
