Disclosed, among other things, is an isolated or recombinant protein that includes extracellular domains of integrin alpha and beta subunits, wherein at least one of the subunits comprises one or more mutations that cause a difference in conformational preference of the protein between bent and extended conformational states, or wherein at least one of the subunits comprises an altered surface feature such that the protein has (1) a difference in conformational preference of the protein between bent and extended states, or (2) a difference in affinity for a cognate ligand under one or more reaction conditions.
