METHOD FOR STRAIGHTENING AND IMMOBILIZING CYCLIC DNA

PROBLEM TO BE SOLVED: To provide a method for straightening and immobilizing a cyclic DNA on a solid substrate. SOLUTION: This method for straightening and immobilizing the cyclic DNA on the solid substrate comprises making a composite single molecular membrane comprising a cationic amphiphilic substance and the cyclic DNA on a gas-water interlace and transferring the composite single molecular membrane to the solid substrate. By the method, the cyclic DNA can efficiently be immobilized on the solid substrate in a cyclic DNA-straightened state in good reproducibility. The immobilized DNA specimen can be used as a specimen for analyzing a base sequence or for analyzing the characteristics of the bond sequence of a protein which is specifically bound to the DNA.
