PURPOSE: A method for preparing an optical active 1,4-benzodioxan-2-carboxylic compounds using enzymatic method is provided, thereby simplifying the preparation method, and improving optical purity and preparation yield. CONSTITUTION: A method for preparing an optical active 1,4-benzodioxan-2-carboxylic compounds using enzymatic method comprises hydrolysis of racemic 1,4-benzodioxan-2-carboxypiperazine using protease derived from a microorganism in the presence of a buffer solution with pH 6-8; adding an organic solvent into the reaction solution to separate the solution into an organic solvent layer and a water-soluble layer; and isolating (S)-1,4-benzodioxan-2-carboxypiperazine from the organic solvent layer or (R)-1,4-benzodioxan-2-carboxylic acid from the water-soluble layer, wherein the microorganism is Aspergillus sp. or Streptomyces sp.; and the hydrolysis temperature is 20 to 40 deg. C.
