PURPOSE: A method for preparing an optically active (R)-2-amino-1-butanol using enzymatic method is provided, thereby removing re-crystallization process, and improving optical purity and preparation yield. CONSTITUTION: A method for preparing an optical active (R)-2-amino-1-butanol using enzymatic method comprises selective hydrolysis of an ester compound of racemic 2-amino-1-butanol using hydrolase derived from a microorganism in the presence of a buffer solution with pH 6-9, wherein the hydrolase is protease or esterase derived from microorganisms, and is powder, liquid or resin immobilized forms; the microorganism is Rhizopus sp. or Candida sp.; the hydrolysis temperature is 20 to 40 deg. C; and ratio of enzyme and substrate in hydrolysis is 1:10 to 10:1.
