<P>PROBLEM TO BE SOLVED: To provide a method for simply and accurately detecting methylated cytocines in a DNA, and to provide an oligonucleotide-immobilizing base plate necessary for the method. <P>SOLUTION: This base plate is used for detecting whether cytosines (Cs) of CpG dinucleotides in a sample DNA which contains a target sequence containing the CpG dinucleotides are methylated or not based on result of hybridization, wherein the hybridization is conducted so that two or more capture oligonucleotides immobilized on the base plate and containing an oligonucleotide having a base sequence complementary to or identical to such a base sequence that all of the Cs other than Cs of all the CpG nucleotides in the target sequence are substituted by Ts (thymines) and another oligonucleotide having another base sequence complementary to or identical to such a base sequence that all of the Cs in the target sequence are substituted by the Ts are hybridized with the sample DNA of which the not-methylated Cs are converted into uracils through deamination or with an amplification product of the DNA. <P>COPYRIGHT: (C)2004,JPO |
