摘要 PURPOSE: A method for modifying DDR2(discoidin domain receptor) tyrosine kinase activity and DDR2 protein having modified kinase activity thereby are provided, thereby treating diseases caused by oxidative stresses and over-activation of DDR2 tyrosine kinase. CONSTITUTION: A method for modifying DDR2(discoidin domain receptor) tyrosine kinase activity comprises the steps of: amplifying cDNA encoding the amino acid sequence containing the DDR2 kinase active site and connecting the amplified sequence to the C-terminal coding region of glutathione-S-transferase(GST) to produce DDR2 kinase-GST fusion gene; cloning the DDR2 kinase-GST fusion gene into an expression vector, expressing the tyrosine kinase active site of DDR2 protein-GST fusion protein in a host cell, and purifying the protein; and adding oxidative stresses to the fusion protein to phosphorylate tyrosine in the tyrosine kinase active site of DDR protein.
