Transcriptional sequencing of DNA, comprising obtaining a nucleic acid transcription product using a modified RNA polymerase having enhanced 3'-deoxynucleotide incorporation ability, template DNA including a promoter sequence for the RNA polymerase, and a substrate for the RNA polymerase, is new. The RNA polymerase is a wild-type RNA polymerase modified by substitution or deletion of one or more amino acid residues.
