METHOD FOR DISCRIMINATING ENDOCRINE DISRUPTORS USING TRANSGENIC YEAST TWO-HYBRID SYSTEM CONTAINING HERαGENE AND CO-ACTIVATOR SRC1 GENE

PURPOSE: A method for discriminating endocrine disruptors using a transgenic yeast two-hybrid system containing hER α gene and co-activator SRC1 gene is provided, thereby rapidly and easily discriminating endocrine disruptors. CONSTITUTION: A method for discriminating endocrine disruptors comprises the steps of: constructing a single-hybrid system containing a human estrogen receptor(hER) α gene and a GAL4 DNA binding domain; constructing an expression vector containing a GLA4 transcription activating domain and a transcription co-activator SRC1; introducing the expression vector Saccharomyces cerevisiae YRG-2 containing a reporter gene group(UASGAL4-TATA-lacZ) on chromosome; culturing the transgenic yeast produced by a two-hybrid system under specific chemical treated condition to express the reporter gene; and monitoring the expression of beta-galactosidase and ligand-dependent transcription activation of estrogen receptor α.
