MUTAGENESIS OF DRUG BINDING REGION OF PROTEINS IN METHODS OF DRUG DISCOVERY

There is a method for assessing the suitability of a drug for therapeutic use. The method involves replicating a nucleic acid sequence encoding a region of a protein to which the drug is capable of binding to provide a library containing mutagenised copies of the nucleic acid sequence. The library is introduced into cells for expression of the library and the cells cultured in the presence of the drug. Clones which are resistant to the drug are selected and the mutations that confer the resistance to the drug are identified. There is also a method for providing a new drug less prone to the mutations on the basis of the mutational data obtained.
