PROBLEM TO BE SOLVED: To provide a method for nucleus inserting operation to a pearl oyster by which nucleus inserting operation is facilitated and defective nucleus insertion and nucleus rejection by influences of pearl oyster body fluid can effectively be prevented in relation to the method for the nucleus inserting operation in order to embed the pearl nucleus into the body of the pearl oyster in culturing the pearl oyster. SOLUTION: This method for nucleus inserting operation to the pearl oyster is characterized as performing the nucleus inserting operation after reducing the body fluid of the pearl oyster. The pearl oyster is dipped in a concentrated seawater solution, a solution at a raised magnesium ion concentration, a solution at a raised concentration of a purgative for livestock or a solution at a raised concentration of bittern to discharge the body fluid of the pearl oyster.
