<P>PROBLEM TO BE SOLVED: To obtain a regulator protein having a nitrilase gene promoter-activating action and originating from Arthrobacter, and to provide the gene. <P>SOLUTION: The gene of the regulator protein having the nitrilase gene promoter-activating action is obtained by extracting and purifying the nitrilase from Arthrobacter sp. NSSC 104, determining the amino acid sequence of the nitrilase, performing a PCR (polymerase chain reaction) using a designed primer for its chromosomal DNA, labeling the amplified DNA with digoxigenin to form a probe, digesting the chromosomal DNA with restriction enzymes, inserting the obtained DNA fragments into a plasmid, transducing Escherichia coli to the plasmid, screening the Escherichia coli by a colony hybridization method to obtain the plasmids into which the DNA fragments containing the nitrilase gene are inserted, determining the base sequences of the DNA fragments, determining the amino acid sequence of a protein capable of being encoded with them from an open reading frame, and then determining the gene of the regulator having the nitrilase gene promoter-activating action by a homology retrieval. <P>COPYRIGHT: (C)2004,JPO |
