<P>PROBLEM TO BE SOLVED: To provide a method and a reagent for increasing efficiency of coamplification reactions such as multiplex PCR (polymerase chain reaction) reaction. <P>SOLUTION: The efficient conditions for multiplex PCR amplification includes using hot start DNA polymerase with a volume exclusion agent in PCR reaction. The positive effect for multiplex PCR is based on, on the one hand, use of a hot start enzyme maximizing PCR efficiency by minimizing undesired nonspecific hybridization and extension of primer and formation of duplex primer. On the other hand, the existence of the volume exclusion agent improves hybridization speed of all reacting primer. By existence of the reacting volume exclusion agent, product yield of a target exhibiting little amplification under a multiplex PCR conditions (defined primer concentration) is much increased. <P>COPYRIGHT: (C)2004,JPO |
