The invention relates to a method for producing S-adenosyl-methionine (SAM) in a production strain comprising the following steps: a) carrying out the concentration process during which the production strain is cultured under conditions, which lead to a propagation of the production strain and which, after optionally adding methionine, lead to a concentration of SAM in the cells of the production strain culture; b) carrying out the SAM fermentation process during which the production strain culture from a) is, while optionally adding methionine, cultured under conditions that lead, in essence, to a concentration of SAM in the cells of the production strain, and; c) optionally isolating the SAM concentrated in the cells of the production strain by decomposing the cells and optionally purifying the isolated SAM. The production strain used in the invention is a prokaryotic or eukaryotic organism, which overexpresses at least one of the following genes: MUP1, MUP3, SAM3, MET28, BAS1, PHO2, GCN4, COQS, SIP18 or one of its homologs in comparison with the wild type production strain or in which at least one copy of at least one of the following genes: MIG1, SPE2, BIO3, ABD1, ERG6, STE14, PEM1, CTM1, HSL7, HMT1, NOP2, DIM1, PET56, ALD6 and SNQ1 is inactivated or which has a combination consisting of these overexpressions and gene inactivations. The invention also relates to a prokaryotic or eukaryotic organism, which overexpresses at least one of the genes selected from the group consisting of: MUP1, MUP3, SAM3, MET28, BAS1, PHO2, GCN4, COQS, SIP18 in comparison with the wild type organism and/or in which at least one copy of at least one of the following genes: MIG1, SAM3, SPE2, BIO3, ABD1, ERG6, STE14, PEM1, CTM1, HSL7, HMT1, NOP2, DIM1, PET56, ALD6 and SNQ1 is inactivated. The invention additionally relates to the use of the aforementioned organisms for producing SAM. |
