BIOSYNTHETIC PROCESS OF TDP-4-KETO-6-DEOXY-D-GLUCOSE AS INTERMEDIATE OF DEOXY SUGAR, AND RECOMBINANT ENZYMES, RECOMBINANT VECTORS AND TRANSFORMANTS THEREFOR

PURPOSE: A biosynthetic process of TDP-4-keto-6-deoxy-D-glucose as an intermediate of deoxy sugar, and recombinant enzymes, recombinant vectors and transformants therefor are provided, thereby cheaply preparing TDP-4-keto-6-deoxy-D-glucose using cheap thymidine monophosphate by one reaction. CONSTITUTION: A biosynthetic process of TDP-4-keto-6-deoxy-D-glucose as an intermediate of deoxy sugar comprises adding thymine monophosphate kinase, acetic acid salt kinase, TDP-glucose synthase and TDP-D-glucose-4,6-dihydratase into a reaction solution containing thymine monophosphate, ATP, acetyl phosphate and glucose-1-phosphate, and reacting them, wherein the thymidine monophosphate is produced from transformed E. coli XL1-Blue/pET15b:tmk(KCTC 10334BP) which is produced by transforming with a recombinant vector pET15b:tmk containing thymine monophosphate gene tmk derived from E. coli K-12; the acetic acid kinase is produced from transformed E. coli XL1-Blue/pET24ma:ack(KCTC 10331BP) which is produced by transforming with a recombinant vector pET24ma:ack containing acetic acid salt kinase gene derived from E. coli K-12; TDP-glucose synthase is produced from transformed E. coli XL1-Blue/pET24ma:rffH(KCTC 10332BP) which is produced by transforming with a recombinant vector pET24ma:rffH containing TDP-glucose synthase gene derived from E. coli K-12; and the TDP-D-glucose-4,6-dihydratase is produced from transformed E. coli XL1-Blue/pRSETB:rfb'B'(KCTC 10333BP) which is produced by transforming with a recombinant vector pRSETB:rfb'B' containing TDP-D-glucose-4,6-dihydratase gene derived from Salmonella typhimurium.
