A door seal structure for sealing between a door opening portion of a vehicle body and a door frame 12(D) of a vehicle door comprises an opening weather strip 40 attached along the door opening portion and a glass run 172 for guiding a door glass 16 held in a glass run holding part 170. The door frame 12(D) has a protrusion 176 protruding inwardly, with respect to the vehicle, of the glass run holding part 170. The seal structure further comprises a cover member 186 for covering the protrusion 176 and concealing it from the interior of the vehicle when the door is closed. The protrusion 176 has a longitudinally extending depression 184 near a protruding wall 182 of the protrusion 176. A first edge 192 of the cover member 186 is connected to the glass run 172 and the other edge 190 of the cover member 186 is seated in the depression 184. The protrusion 176 may have a second longitudinally extending depression 184 near an inner side wall 178 of the glass run 172 in which the first edge 192 of the cover member 186 is seated.
