An N-6 substituted 7-deazapurine or pharmaceutically acceptable salts thereof has the formula (I) wherein: R1 and R2 are independently hydrogen or a substituted alkyl, substituted cycloalkyl or optionally substituted aryl wherein only one of R1 and R2 may be hydrogen and when the substituted alkyl is -(CH2)-phenyl or -(CH2-CH2)-phenyl then the phenyl is substituted or R1 and R2 together form an optionally substituted heterocyclic ring, R3 is optionally substituted aryl; R4 is hydrogen or unsubstituted alkyl or optionally substituted aryl and R5 and R6 are independently halogen, hydrogen or an optionally substituted alkyl, aryl, or alkylaryl or R4 and R5 or R5 and R6 together form an optionally substituted heterocyclic or carbocyclic ring. A deazapurine has the formula II wherein: X is N or CR6; R1 and R2 are independently hydrogen or optionally substituted alkoxy, aminoalkyl, alkyl, aryl, or alkylaryl wherein only one of R1 and R2 may be hydrogen or R1 and R2 together form an optionally substituted heterocyclic ring; R3 is optionally substituted arylalkyl, heteroaryl or aryl; R4 is hydrogen; L is hydrogen, optionally substituted alkyl, or R4 and L together form an optionally substituted heterocyclic or carbocyclic ring; R6 is hydrogen, optionally substituted alkyl, or halogen; Q is CH2, O, S, or NR7; R7 is hydrogen or optionally substituted alkyl; and W is optionally substituted alkyl, cycloalkyl, alkynyl, aryl, arylalkyl, biaryl, heteroaryl, substituted carbonyl, substituted thiocarbonyl, or substituted sulphonyl. The compounds are useful for treating a disease or condition associated with increased levels of adenosine in a subject wherein the disease or condition is a central nervous system disorder, a cardiovascular disorder, a renal disorder, an inflammatory disorder, an allergic disorder, a gastrointestinal disorder or a respiratory disorder.
