The present invention is directed to a method of detecting a T cell response induced by smallpox vaccines or Orthopoxvirus vector vaccines in a mammal and to immunogenic Orthopoxvirus antigens and the nucleic acids encoding same. The present invention further relates to pharmaceutical compositions and their use in the protective immunization against a smallpox infection.
