Methods for purifying and detecting double stranded DNA target sequences by triple helix interaction.

Purifying a double-stranded (ds) DNA molecule (I) comprising contact with a third DNA strand (II) that interacts with a target sequence (TS) in (I) to form a triple helix, is new. TS has the formula 5'-(R) n-(N) t-(R') m-3' R and R' : nucleotide sequences comprising purine bases only; n and m : integers below 9, but totaling over 5; N : nucleotide sequence comprising both purine and pyrimidine bases; t : integer below 8 Independent claims are also included for the following: (a) (I) purified by the new method; and (b) detecting (I) by reaction with labeled (II).
