FIELD: biotechnology. SUBSTANCE: invention relates to methods for preparing and purification of genetic-engineering preparations of human interferon. Method involves culturing cells of microorganism Pseudomonas putida, cells lysis with their simultaneous disruption, treatment of cells with polyethyleneimine, separation of nucleic acids, proteins desalting with ammonium sulfate, three-stage chromatography on cation-exchange hydrophobic sorbent Solosa KG, anion-exchange resin Spherocell-QAE and hydrophobic sorbent Spherocell LP-M followed by concentrating by methods of ultrafiltration and gel-filtration on Sephadex G-100. The proposed purification schedule provides enhancing the yield of interferon up to 30%. EFFECT: improved preparing method, enhanced yield of interferon. 1 ex |
