PROBLEM TO BE SOLVED: To provide a new tricyclic condensed heterocyclic compound and its production method and use. SOLUTION: A pharmaceutical composition comprises a compound represented by formula (1), its optical isomer and its conjugate, or their pharmaceutically permissible salts (in formula (1), X is CH, CH<SB>2</SB>, CHR or CRR'; Y is CH, CH<SB>2</SB>or C=O; Z is selected from O, S, S=O and SO<SB>2</SB>; U is C or N; R<SB>1</SB>-R<SB>4</SB>are each independently a hydrogen atom, OR, SR, an aromatic ring, a substituted aromatic ring or a heterocyclic ring; at least one of R<SB>5</SB>-R<SB>8</SB>is, for example, OH and the others are each independently a hydrogen atom or OH; and R and R' are each a lower alkyl group or a substituted lower alkyl group). These compounds have a wide range of the following pharmacological effects: an excellent effect on relaxant of the tracheae smooth muscle, effects on suppressing airway hypersensitivity and on suppressing invasion of inflammatory cells into the airway, and they also have a high safety.
