<P>PROBLEM TO BE SOLVED: To provide a dumbbel type nonviral vector improved in expression efficiency. <P>SOLUTION: The dumbbel type vector is obtained by linearizing a cyclic plasmid DNA and capping the both ends of the linearized DNA with an oligonucleotide forming a thermally/thermodynamically stable loop. The thermally/thermodynamically stable loop exhibits a resistance to exonuclease (catabolic enzyme) and DNA arriving a nuclei without being decomposed expresses a gene. The vector is represented by the general formula A-R-B [wherein A and B may be each same or different and A and B are each a loop composed of a single chain oligonucleotide containing the base sequence gaa or gnaa (n is a, g, c or t) and having ≤6 base number and a double-stranded straight chain DNA having 4-20 kb base logarithm; R is a double-stranded straight chain DNA having 1-10 kb base logarithm and comprises a promoter, a foreign gene and poly A-added signal]. The vector exhibits expression efficiency (transcription efficiency) which is 5-10 times higher than that of the cyclic DNA. <P>COPYRIGHT: (C)2004,JPO |
