<p>PURPOSE:To obtain a new monoclonal antibody belonging to IgGlkappa class, specifically bindable to 18-hydroxycortisol, thus capable of quickly determining 18- hydroxycortisol in the body fluid and useful for diagnosing primary aldosteronism. CONSTITUTION:18-hydroxycortisol is bound to a carrier protein (e.g. keyhole limpet hemocyanin), the product is then mixed with Freund's complete adjuvant and administered to mice or rats to immunize them. Three days after the final immunization, the splenocytes are taken from them and fused with mouse myeloma cells in the presence of polyethylene glycol followed by selective culture in a HAT medium to obtain hybridoma. The hybridoma is then screened to select a clone having ability to produce an antibody against 18- hydroxycortisol, and a cloning is performed by limiting dilution method; the resultant monoclonal hybridoma is cultured to obtain the objective monoclonal antibody belonging to IgGlkappa class and specific to 18-hydroxycortisol.</p> |
