Preparing a synthetic polypeptide (I) with at least one detectable function, is new. Preparing a synthetic polypeptide (I) with at least one detectable function comprises: (1) comparing amino acid (aa) sequences of at least 3 known polypeptides that have at least one function in common; (2) equalizing the position of invariant aa, or similar regions of aa, by mutual alignment and/or introduction of gaps, in one or more sequences, to produce a correspondence between the positions of the invariant aa and similar sequences; (3) establishing an 'average sequence' (AS) over at least a major part of the total length of the aa sequence, by selecting the commonest aa at each position or, where several aa have equal frequencies, selecting one of these according to preselected criteria, with gaps being treated as aa; (4) designing, then synthesizing, a nucleic acid sequence (II) that encodes AS; and (5) linking (II) to a promoter for expression in a suitable system. Independent claims are also included for the following: (1) (I) as new compounds; (2) isolated or synthetic nucleic acids (IIa) that encode (I); (3) a vector for expressing (I) that contains (IIa); and (4) a cell, or kit, that contains (I), (IIa) and/or the vector of (3).
