The invention relates to a biochip comprising a substrate having a main reflective surface. The inventive biochip is characterised in that the main surface is provided with areas that are made sensitive to fluorescence detection by a transparent layer of thickness (2k+1) lambda /4, wherein k is a positive integer or zero, lambda denotes a wavelength that is included between a fluorescence excitation wavelength lambda o and a fluorescence emission wavelength lambda 1. Said biochip can be produced by depositing a transparent layer of thickness m lambda '/2 on a substrate and creating pits or bumps of thickness (2k+1) lambda /4 in said layer in order to form sensitive areas.
