PURPOSE: A process for preparing 9-£4-acetoxy-3- (acetoxymethyl)but-1-yl|- 2-aminopurine is provided, thereby decreasing the production of by-products and preparing purine derivative in higher selectivity under mild condition. CONSTITUTION: A process for preparing 9-£4-acetoxy-3- (acetoxymethyl)but-1-yl|- 2-aminopurine of the formula(I) comprises reacting a compound of the formula(XXI) with a compound of the formula(III) under a polar organic solvent, wherein the polar organic solvent is selected from N,N-dimethylformamide, diphenylsulfoxide and C1-C3 lower alcoholic solvent; the reaction is carried out at 0 to 100 deg. C; and the compound of the formula(XXI) is prepared by reacting a compound of the formula(II) with a compound of the formula(IV) under a polar organic solvent selected from N,N-dimethylformamide, diphenylsulfoxide and C1-C3 lower alcoholic solvent at 0 to 100 deg. C.
