Piece of gel is separated from a gel section (i.e. a gel cut-out) with an outline that would suite contour of gel piece holder, and is positioned into gel holder by: e.g. separating from gel a gel section (27) having edge contour that corresponds precisely or approximately to contour of a chamber for receiving gel section in gel holder (21); and covering the gel section with equilibrating fluid (31).
