<P>PROBLEM TO BE SOLVED: To provide a method for producing recombinant fibrinogen, by which the recombinant fibrinogen having the perfect molecular structure can stably and massively be produced by a genetic engineering method using a Pichia yeast. <P>SOLUTION: This method for producing the recombinant fibrinogen having the perfect molecular structure of (Aα-Bβ-γ)<SB>2</SB>includes (1) a process for connecting a promoter capable of controlling the expression of a different kind of protein in the Pichia yeast, a fibrinogen-originated signal sequence, and a gene encoding the Aα chain, Bβ chain and γ chain of the fibrinogen to each other to prepare the expression plasmid, (2) a process for transducing the plasmid into the Pichia yeast to prepare the transformant, (3) a process for culturing the transformant in the condition of pH 5 to 8 to produce the fibrinogen, and (4) a process for purifying the fibrinogen recovered from the cultured supernatant. <P>COPYRIGHT: (C)2004,JPO |
